Is is possible to change the word 'subscribe' to 'join' on the form?

Yes, this is possible, that label is being controlled by a language file. Edit the following file:

/language/en-GB/en-GB.com_rsmembership.ini

and look for

COM_RSMEMBERSHIP_SUBSCRIBE="Subscribe"

Simply replace "Subscribe" with "Join" and save the file.

Hi - will this edit be preserved if the extension is updated?

i.e. can the ini file be overridden?

I just saw this. Yes, you can use the Joomla Language Overrides feature. In admin go to Extensions > Language Manager > Overrides and search for the term you want to edit.
